Night Passage. Here's a longer clip of the scene. Note that, while he may or may not be playing the accordion heard on the soundtrack (I suspect not), it was obviously recorded separately--his fingers are not quite playing the same song.
Bonus appearance at the end from Ellen Corby, perhaps better-known as playing the grandmother on The Waltons... which had some nice accordion work on its soundtrack too.
Edited to add: A-ha! Per the notes at the end of the Wikipedia article I linked, Jimmy Stewart's "accordion playing was rerecorded by a professional during post-production".
