If there are mike connectors fitted into the housing of an accordion, say one at the top of the treble keyboard and one at the bottom, and these are not used, can they have any effect at all on the acoustic sound? After all flaps are fitted on some accordions which when opened increase the volume ( I believe ).
e.g. vibration, deadening, etc

Well, depends on where the hole is drilled. If it is drilled INSIDE if the bellows cavity, it would have a huge effect. If it is outside of the bellows cavity, then the only effect would be cosmetic damage.

Changes to the grille can affect the sound of the accordion. Two holes, one at the top and one at the bottom, where a mic connector used to be, will not have any effect (other than cosmetic). Larger changes will affect the sound. If you remove the grille completely the accordion you will hear how different the sound is. Some accordions allow to vary the sound by means of valves (sometimes called saxophone valves) or a movable mesh screen, for instance on the Hohner Gola and the old Atlantic IV (not the N version).
Some things you add or remove "under the grille" may cause vibrations or other noises and these can sometimes be hard to identify. (For instance, the grille on my Pigini basson caused a vibration where the cutout for the register switches is, and a piece of felt solved that.) But the two mic holes are unlikely to have any ill effect.
